If you missed the chance to edit your screenshot, you can still do so in the Photos app. Tap your screenshot, select Edit at the top, tap the three dots at the top, and then choose Markup to open the editor.
If you don't want to use AssistiveTouch anymore, you can return to Settings > Accessibility > Touch > AssistiveTouch and tap the switch to turn it off.
